Resumo: Додаткові функції протеїнів казеїнового комплексу молока проявляються через біоактивні продукти їх протеолізу – пептиди. Такі пептиди можуть впливати на функції травної, серцево-судинної, нервової та імунної систем організму. Серед біоактивних казеїнових пептидів одними з найважливіших є фосфопептиди. Їх основна функція – забезпечення транспортування і засвоєння організмом іонів двовалентних металів. Також вони можуть проявляти інші види біологічної дії. Очевидно, що такими корисними властивостями володіють природні або аналогічні до природних фосфопептиди. Такі пептиди можуть утворюватися в умовах, які мають місце у шлунково-кишковому тракті. При проведенні протеолізу в таких умовах важливим доказом ідентичності отриманих фосфопептидів природним фосфопептидам є молекулярно-масовий розподіл. У зв’язку з цим метою роботи була характеристика молекулярно-масового розподілу казеїнових фосфопептидів, отриманих з відтворенням умов природного протеолізу. Казеїнові фосфопептиди було отримано в результаті протеолізу загального фосфопротеїну коров’ячого молока в умовах, які мають місце у шлунково-кишковому тракті. Молекулярно-масовий розподіл виділених фосфопептидів визначали за допомогою гель-фільтрації. При цьому було використано три типи сефадексів: G-10, G-15 і G-25. Склад і гомогенність фосфопротеїнових субстратів аналізували електрофоретично в лужній системі поліакриламідного гелю. В результаті було встановлено, що препарат казеїнових фосфопептидів, отриманий в умовах, близьких до умов протеолізу в шлунково-кишковому тракті, містить 52 % фосфопептидів з молекулярними масами від 700 до 1500 Да. В цей діапазон молекулярних мас попадає більшість відомих казеїнових фосфопептидів коров’ячого молока. Отримані результати були використані при розробці біотехнології виділення природних казеїнових фосфопептидів.
The additional functions of proteins of the casein complex of milk are manifested through the bioactive products of their proteolysis — peptides. Such peptides may affect on the functions of the digestive, cardiovascular, nervous and immune systems of the body. Among the bioactive casein peptides, one of the most important are phosphopeptides. Their main function is to transport provision and absorption of ions of bivalent metals by the body. They can also exhibit other biological effects. Obviously, such beneficial properties are natural or similar to natural phosphopeptides. Such peptides may be formed in conditions that occur in the gastrointestinal tract. When conducting proteolysis in such conditions, an important proof of the identity of the resulting phosphopeptides with natural ones is the molecular-mass distribution. In this regard, the purpose of the work was to characterize the molecular weight distribution of casein phosphopeptides obtained with the reproduction of the conditions of natural proteolysis. Casein phosphopeptides were obtained as a result of proteolysis of the total phosphoproteins of cow's milk in conditions that occur in the gastrointestinal tract. The molecular weight distribution of the isolated phosphopeptides was determined using gel filtration. In this case, three types of sephadexes were used: G-10, G-15 and G-25. The composition and homogeneity of phosphoprotein substrates were analyzed electrophoretically in an alkaline system of the polyacrylamide gel. As a result of the fulfilled experiments, it was found that the prepared casein phosphopeptides, in the conditions close to proteolytic ones in the gastrointestinal tract, contained 52% of phosphopeptides with molecular weights of 700 to 1500 Da. In this range of the molecular masses, most of the known casein phosphopeptides of cow's milk are found. The obtained results were used during development of biotechnology for the separation of the natural casein phosphopeptides.
